Biochemical properties and biological functions of the enzyme rhodanese in domestic animals

The enzyme rhodanese (thiosulfate: cyanide sulfurtransferase) is a ubiquitous enzyme and its activity ispresent in all living organisms. Many functions including cyanide detoxification, formation of iron-sulfurcenters and participation in energy metabolism have been attributed to this enzyme. The enzyme catalyzesthe transfer of a sulfur atom from sulfane containing compounds (such as thiosulfat...

Optimization of Liposomal Lipid Composition for a New, Reactive Sulfur Donor, and In Vivo Efficacy Studies on Mice to Antagonize Cyanide Intoxication

Present studies have focused on a novel cyanide antidotal system, on the coencapsulation of a new sulfur donor DTO with rhodanese within sterically stabilized liposomes. The optimal lipid composition for coencapsulation of DTO with rhodanese is the combination of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine, cholesterol, cationic lipid (DOTAP), and 1,2-dipalmitoyl-sn-glycero-3-phosphoethano...

Mechanism of rhodanese action: isotopic tracer studies.

In a previous publication (2) a double displacement mechanism for the catalytic activity of rhodanese (thiosulfate cyanide sulfurtransferase) was proposed on the basis of polarographic evidence. In addition it was concluded from the polarographic data that crystahine beef liver rhodanese is an enzyme-substrate intermediate containing two activated substrate sulfur atoms per molecule. This commu...

Molecular cloning of rhodanese gene from soil metagenome of cold desert of North-West Himalayas: sequence and structural features of the rhodanese enzyme

Rhodanese is a multifunctional, sulfur transferase that catalyzes the detoxification of cyanide by sulphuration in a double displacement (ping pong) mechanistic reaction. In the present study, small-insert metagenomic library from soil sample collected from Ladakh (3,000-3,600 m.a.s.l) in northwestern Himalayas, India was constructed. Function-driven screening of ~8,500 colonies led to the isol...

The isolation of rhodanese from Pseudomonas aeruginosa by affinity chromatography.

Rhodanese (thiosulphate : cyanide sulphur-transferase; EC 2 . 8 . I . I) was originally ,described by Lang (1933). The enzyme catalyses the formation of thiocyanate from cyanide and thiosulphate according to the reaction : S20!+ CN-+ SCN+ SO:-. Rhodanese activity has been demonstrated in most mammalian tissues, with the greatest activity present in liver and kidney. The enzyme has been purified...
